Product Description:
Polyfectine is a nanoparticle-based transfection reagent optimized for a wide spectrum of mammalian cells including dividing cells and non-dividing cells. Compared to lipofectamine, Polyfectine demonstrates very low cytotoxicity. It can be used for DNA or SiRNA transfection. The applicable ratio of DNA (or SiRNA): Polyfectine ranges from 1:2 to 1:3.
Standard Transfection Procedure in 6-well plates:
1. Adherent cells: One day before transfection, plate 3-5x 105 cells in 2ml of growth medium without antibiotics so that cells will be 90-95% confluent at the time of transfection.
2. Suspension cells: Just prior to preparing complexes, plate 1-2 x 106 cells in 2ml of growth medium without antibiotics.
3. For each transfection sample, prepare complexes as follows:
a. Dilute 4ug of the DNA in 200 μl of Opti-MEM® I Reduced Serum Medium without serum (or other medium without serum). Mix gently.
b. Add 12 μl of the Polyfectine to each tube containing diluted DNA. Mix gently and incubate for 15 minutes at room temperature.
3. Add the 220 μl of complexes to each well containing cells and medium. Mix gently by rocking the plate back and forth.
4. Incubate cells at 37°C in a CO2 incubator for 18-48 hours prior to testing for transgene expression. Medium may be changed after 4-6 hours.
Storage
The kit can be stored for 3 months at room temperature, for 1 year at 4℃ and over 1 year at -20℃.
