E3 ubiquitin-protein ligase Mdm2 (MDM2) is a E3 ubiquitin-protein ligase that mediates ubiquitination of p53/TP53, leading to its degradation by the proteasome. Inhibits p53/TP53- and p73/TP73-mediated cell cycle arrest and apoptosis by binding its transcriptional activation domain. Also acts as a ubiquitin ligase E3 toward itself and ARRB1. Permits the nuclear export of p53/TP53. Promotes proteasome-dependent ubiquitin-independent degradation of retinoblastoma RB1 protein. Inhibits DAXX-mediated apoptosis by inducing its ubiquitination and degradation.
Canine E3 ubiquitin-protein ligase Mdm2 (MDM2) ELISA Kit employs a two-site sandwich ELISA to quantitate MDM2 in samples. An antibody specific for MDM2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMDM2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MDM2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MDM2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
