Cleaved Caspase-8 (Asp374) is a key protein involved in the extrinsic apoptosis pathway, which is activated by death receptors such as Fas (CD95) and tumor necrosis factor receptor 1 (TNFR1). When cleaved at Asp374, it generates active fragments, including the p18 subunit, which can further activate downstream effector caspases like caspase-3, -6, and -7, ultimately leading to apoptosis. This protein is also implicated in regulating inflammatory necrotic cell death pathways such as necroptosis and pyroptosis.
