Receptor tyrosine kinases interact with SRC domains to enzymatically activate PLCγ1 via phosphorylation at tyrosine 783. Upon activation PLCG1 leads to the translocation of Ras guanine nucleotide exchange factor RasGRP1 to the Golgi apparatus, activating Ras. PLCG1 is also a primary substrate of heparin-binding growth factor 1 (acidic fibroblast growth factor)-activated tyrosine kinases.
