TRPM3 belongs to the family of transient receptor potential (TRP) channels. TRP channels are cation-selective channels important for cellular calcium signaling and homeostasis. The protein encoded by this gene mediates calcium entry, and this entry is potentiated by calcium store depletion. Alternatively spliced transcript variants encoding different isoforms have been -identified. TRPM3 was shown to be activated by the neurosteroid pregnenolone sulphate in hepatocytes. The activation causes calcium influx and subsequent insulin release, therefore it is suggested that TRPM3 modulates glucose homeostasis.TRPM3 expression in the cytoplasm of collecting tubular epithelium. Immunofluorescence and confocal microscopy showed that HA-tagged full-length TRPM3 localizes to the plasmalemmal compartment.
Mouse Transient receptor potential cation channel subfamily M member 2 (TRPM2) ELISA Kit employs a two-site sandwich ELISA to quantitate TRPM2 in samples. An antibody specific for TRPM2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTRPM2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TRPM2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TRPM2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
