Delta-sarcoglycan isone of the four known components of the sarcoglycan complex, which is a subcomplex of the dystrophin-glycoprotein complex (DGC). The sarcoglycans are a family of transmembrane proteins involved in the protein complex responsible for connecting the muscle fibre cytoskeleton to the extracellular matrix, preventing damage the muscle fibre sarcolemma through shearing forces. This protein is expressed most abundantly in skeletal and cardiac muscle. The mutations in this gene have been associated with autosomal recessive limb-girdle muscular dystrophy and dilated cardiomyopathy. Alternatively spliced transcript variants encoding distinct isoforms have been observed.In melanocytic cells SGCD gene expression may be regulated by MITF.
Human Delta-sarcoglycan (SGCD) ELISA Kit employs a two-site sandwich ELISA to quantitate SGCD in samples. An antibody specific for SGCD has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySGCD present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SGCD is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SGCD bound in the initial step. The color development is stopped and the intensity of the color is measured.
