Stathmin 1/oncoprotein 18, also known as STMN1, is a highly conserved 17 kDa protein. Its function as an important regulatory protein of microtubule dynamics has been well characterized. Eukaryotic microtubules are one of three major components of the cell’s cytoskeleton. They are highly dynamic structures that continuously alternate between assembly and disassembly. Stathmin performs an important function in regulating rapid microtubule remodeling of the cytoskeleton in response to the cell’s needs. Microtubules are cylindrical polymers of α,β-tubulin. Their assembly is in part determined by the concentration of free tubulin in the cytoplasm.At low concentrations of free tubulin, the growth rate at the microtubule ends is slowed and results in an increased rate of depolymerization (disassembly).
Human Stathmin-1 (STMN1) ELISA Kit employs a two-site sandwich ELISA to quantitate STMN1 in samples. An antibody specific for STMN1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySTMN1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for STMN1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of STMN1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
