TM4SF18 also known as AAB5 and L6D, was cloned from human umbilical vein endothelial cells. It shares approximately 40 - 50% amino acid sequence identity with L6, IL-TMP, and TM4SF5, a group of proteins with limited homology to the tetraspanin superfamily.TM4SF18 belongs to the L6 tetraspanin family, L6, IL-TMP, and TM4SF5 are cell surface proteins predicted to have four transmembrane domains. Previous sequence analysis led to their assignment as members of the tetraspanin superfamily.A new sequence (L6D) that is strikingly similar to L6, IL-TMP, and TM4SF5. Analyses of these four sequences indicate that they are not significantly related to genuine tetraspanins, but instead constitute their own L6 superfamily.
Human Transmembrane 4 L6 family member 18 (TM4SF18) ELISA Kit employs a two-site sandwich ELISA to quantitate TM4SF18 in samples. An antibody specific for TM4SF18 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTM4SF18 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TM4SF18 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TM4SF18 bound in the initial step. The color development is stopped and the intensity of the color is measured.
