WAP four-disulfide core domain protein 2 is a protein that is a member of the WFDC domain family. The WFDC domain, or WAP Signature motif, contains eight cysteines forming four disulfide bonds at the core of the protein, and functions as a protease inhibitor in many family members. This gene is expressed in pulmonary epithelial cells, and was also found to be expressed in some ovarian cancers. The encoded protein is a small secretory protein, which may be involved in sperm maturation. Expressed in a number of normal tissues, including male reproductive system, regions of the respiratory tract and nasopharynx. Highly expressed in a number of tumors cells lines, such ovarian, colon, breast, lung and renal cells lines. Initially described as being exclusively transcribed in the epididymis.
Human Epididymal protein 4 (HE4) ELISA Kit employs a two-site sandwich ELISA to quantitate HE4 in samples. An antibody specific for HE4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyHE4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for HE4 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of HE4 bound in the initial step. The color development is stopped and the intensity of the color is measured.
