Exportin-1 mediates leucine-rich nuclear export signal (NES)-dependent protein transport. Exportin 1 specifically inhibits the nuclear export of Rev and U snRNAs. It is involved in the control of several cellular processes by controlling the localization of cyclin B, MPAK, and MAPKAP kinase 2.This protein also regulates NFAT and AP-1.The directionality of nuclear export is thought to be conferred by an asymmetric distribution of the GTP- and GDP-bound forms of Ran between the cytoplasm and nucleus. Involved in U3 snoRNA transport from Cajal bodies to nucleoli. Binds to late precursor U3 snoRNA bearing a TMG cap. Several viruses, among them HIV-1, HTLV-1 and influenza A use it to export their unspliced or incompletely spliced RNAs out of the nucleus.
Human Exportin-1 (XPO1) ELISA Kit employs a two-site sandwich ELISA to quantitate XPO1 in samples. An antibody specific for XPO1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyXPO1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for XPO1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of XPO1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
