MYO6 encodes a reverse-direction motor protein that moves toward the minus end of actin filaments and plays a role in intracellular vesicle and organelle transport. Myosin-VI consists of a motor domain containing an ATP- and an actin-binding site and a globular tail which interacts with other proteins. Myosin-VI maintains the structural integrity of inner ear hair cells and mutations in this gene cause non-syndromic autosomal dominant and recessive hearing loss. Alternative splicing results in multiple transcript variants encoding distinct isoforms.
Chicken Myosin-VI (MYO6) ELISA Kit employs a two-site sandwich ELISA to quantitate MYO6 in samples. An antibody specific for MYO6 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMYO6 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MYO6 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MYO6 bound in the initial step. The color development is stopped and the intensity of the color is measured.
