Anti-B Affinity Chromatography Resins is used for the removal of anti-B agglutinins in blood products. The chromatographic operation typically includes steps such as equilibration, sample loading, washing, elution, and cleaning.
The specific operating methods are as follows:
Equilibration: Equilibrate the column with 5-10CV of equilibration buffer (e.g., 20mM HAc-NaAc, pH5.5) at a flow rate not exceeding the maximum pressure limit, until the conductivity and pH of the effluent remain constant (consistent with the equilibration buffer).
Sample Loading: The sample loading buffer should be as consistent as possible with the equilibration buffer. To avoid clogging the column, the sample solution should be centrifuged or microfiltered (0.45μm). Start collecting the flow-through after peak elution.
Washing: Continue washing with equilibration buffer after sample loading until the UV absorbance drops to baseline, and stop collecting the flow-through to obtain the sample with anti-B agglutinins removed.
Elution: Elute and collect the effluent using elution buffer (typically by increasing NaCl concentration, e.g., 20mM HAc-NaAc + 1M NaCl, pH5.5).
Cleaning: After use, the anti-B affinity resins needs to be cleaned. The recommended cleaning steps are as follows:(1) Clean the media with 0.5M NaOH for 30 minutes;(2) Re-equilibrate with equilibration buffer for the next purification cycle, or store in 20% ethanol after adjusting the pH to neutral.
Storage: Store at 2-10°C in 20% ethanol; the unpacked resins can be stored at 2-10°C after rinsing with 20% ethanol.
Other considerations: During use and storage of the column, avoid running it dry or having loose seals to prevent air bubbles from entering.
