CD138 IHCstart Kit: A Comprehensive Solution for Precise Immunohistochemical Detection
Designed for researchers and lab technicians specializing in immunohistochemistry (IHC) studies, the CD138 IHCstart Kit (Catalog No.: S0I0004) from Starter-Bio is a reliable tool tailored for the specific detection of Syndecan-1 (CD138)—a critical marker closely linked to epithelia, plasma cells, and tumor progression research. This all-inclusive kit streamlines IHC workflows by providing pre-optimized components, ensuring consistent and accurate staining results for formalin-fixed paraffin-embedded (FFPE) tissues (IHC-P application), with cross-reactivity specifically validated for human (Hu) samples.

Each kit contains two core components essential for 50 IHC slides: 25μl of CD138 Recombinant Rabbit mAb (SDT-R120, Catalog No.: S0B2159), which also supports immunocytochemistry (ICC), and 5ml of Anti-Rabbit and Mouse HRP-DAB IHC Detection Kit (2-step, Catalog No.: S0C2011) for efficient signal amplification. The primary antibody, when used at the recommended dilution of 1:1000 for IHC, is supplied in excess to accommodate flexible experimental needs. The kit’s components are formulated in a stable buffer system (PBS with 40% Glycerol, 0.05% BSA, and 0.03% Proclin 300) and require specific storage conditions—primary antibody at -20°C (12-month shelf life from receipt/reconstitution) and detection kit at 2-8°C (12-month shelf life)—to maintain optimal performance.

Beyond its practical components, the kit addresses key research needs related to CD138: as a marker involved in cell adhesion, proliferation, and tumor invasiveness, CD138’s abnormal expression (e.g., loss of membrane staining or gain of cytoplasmic/nuclear staining) and stromal localization are linked to poor prognosis in certain cancers (PMID: 33921767). This kit enables researchers to efficiently investigate such expression patterns, supporting studies in oncology, cell biology, and translational research.

    Background

Syndecan-1 (CD138) is mostly restricted to epithelia, and bears heparan sulfate chains that are capable of interacting with a large array of polypeptides, including extracellular matrix components and potent mediators of proliferation, adhesion and migration. For this reason, it has been studied extensively with respect to carcinomas and tumor progression. Frequently, but not always, syndecan-1 levels decrease as tumor grade, stage and invasiveness and dedifferentiation increase. However, in some tumors, levels of syndecan-1 increase, but the characterization of its distribution is relevant. There can be loss of membrane staining, but acquisition of cytoplasmic and/or nuclear staining that is abnormal. Moreover, the appearance of syndecan-1 in the tumor stroma, either associated with its cellular component or the collagenous matrix, is nearly always a sign of poor prognosis [PMID: 33921767].The reagent can complete 50 slides for IHC experiment and the primary antibody is excessive.
