Intended Use
Ready to use culture medium based assay for the qualitative detection of microorganisms in body fluids
like blood, ascites, cerebrospinal fluid, etc, offering isolated strains to carry out susceptibility tests.
Introduction
Blood culture is one of the most important and critical procedures performed in the microbiology
laboratory. Since blood is normally sterile, the isolation and identification of an organism has great
diagnostic significance. Mostly used to diagnosis microorganisms specially for some fastidious
organisms such as Haemophilus influenzae、 neisseria meningitides、 streptococcus pneumonia、
bacterium burgeri、 actinobacillus、 cardiobacterium hominis、 kingella. It is important to diagnosis
infection bacteremia and septicemia.
Principle
Biphase system is simple to separation cultivation, non-contaminate, easy to observe the isolated
colony on the agar. The isolated colony can use to antibiotics susceptibility test and strains
identification.
Kit Components
Blood culture has Biphase blood culture and Anaerobic blood culture two types.
Biphase blood culture: peptone、 beef extract、 hemin、 yeast、 SPS、 autoclaving
Anaerobic blood culture: peptone、 beef extract、 hemin、 yeast、 SPS、 cysteine hydrochloride、
autoclaving
Storage And Validity
Store all components at 2-25 ℃. Avoid strong light.
The validity duration is 18 months
Specimen Collection
Blood or ascites, sterile operation, can inject the sample in the bottle directly, no need special operate.
Materials Required But Not Provided
Incubator (35-37 ℃)
Procedure
Read the entire procedure carefully prior to testing.
1、 Check if the bottle is contaminated or broken.
2、 Prepare and label the appropriate blood culture bottle
3、 Do not unscrew cap. Remove the plastic top of the screw cap on the blood culture bottle.
4、 Disinfect the visible part of the rubber plug with iodine or ethyl alcohol (70%) and allow drying.
5、 Collect approximately 5-10 ml of adult patients’ blood , or 1-5 ml of infant patients’ blood.
6、 Inject blood, ascites or cerebrospinal fluid into the culture bottle, after being collected with
asepticoperations. Then disinfect the rubber plug again and replace the plastic top.
7、After the inoculation, lean the bottle several times, and incubate it,erectly at （36±1）℃ for 24 hours.
Interpretation Of Results
1、Report the positive results if turbidity in the culture broth, hemolysis of blood corpuscles, air bubbles
or colonies is observed. Carry out susceptibility tests and strain identification on positive samples.
2、 Soak agar state into broth every day and keep on incubating the bottle erectly if those phenomena do
not occur, until the 7th day.
3、 If the above mentioned phenomena still do not occur after 7 days of culturing, report a negative
result after confirm with microscopy and blind passage.
Limitation Of The Test
The basement to diagnostic bacteria is collecting the right specimens, should be confirm with
microscopy and biochemical identification once diagnostic bacteria.
Precautions
1、 For in-vitro diagnostic professional use only.
2、 Blood anticoagulant has already been applied in the culture bottle, so do not add more.
3、 Should be incubated immediately when collect the specimen in case the strains be alive.
