p-ANCA, or MPO-ANCA, or Perinuclear Anti-Neutrophil Cytoplasmic Antibodies, show a perinuclear staining pattern. This pattern occurs because the vast majority of the antigens targeted by ANCAs are highly cationic (Positively charged) at pH 7.00. During ethanol (pH ~7.0 in water) fixation, antigens which are more cationic migrate and localize around the nucleus, attracted by its negatively charged DNA content. Antibody staining therefore results in fluorescence of the region around the nucleus.
Rat Perinuclear anti-neutrophil cytoplasmic antibody (pANCA) ELISA Kit employs the competitive enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with an antibody specific to pANCA. Standards or samples are then added to the appropriate microtiter plate wells with a Horseradish Peroxidase (HRP)-conjugated pANCA and incubated. The competitive inhibition reaction is launched between with HRP labeled pANCA and unlabeled pANCA with the antibody. A substrate solution is added to the wells and the color develops in opposite to the amount of pANCA in the sample. The color development is stopped and the intensity of the color is measured.
