Cyp39a1 encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This endoplasmic reticulum protein is involved in the conversion of cholesterol to bile acids. Its substrates include the oxysterols 25-hydroxycholesterol, 27-hydroxycholesterol and 24-hydroxycholesterol. Alternate splicing results in multiple transcript variants.
Mouse 24-hydroxycholesterol 7-alpha-hydroxylase (CYP39A1) ELISA Kit employs a two-site sandwich ELISA to quantitate CYP39A1 in samples. An antibody specific for CYP39A1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCYP39A1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CYP39A1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CYP39A1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
