The protein encoded by MKRN3 contains a RING (C3HC4) zinc finger motif and several C3H zinc finger motifs. MKRN3 is intronless and imprinted, with expression only from the paternal allele. Disruption of the imprinting at this locus may contribute to Prader-Willi syndrome. An antisense RNA of unknown function has been found overlapping MKRN3.

After removing any unbound substances, a biotin-conjugated antibody specific for MKRN3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MKRN3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
