SDHC encodes one of four nuclear-encoded subunits that comprise succinate dehydrogenase, also known as mitochondrial complex II, a key enzyme complex of the tricarboxylic acid cycle and aerobic respiratory chains of mitochondria. The encoded protein is one of two integral membrane proteins that anchor other subunits of the complex, which form the catalytic core, to the inner mitochondrial membrane. Several related pseudogenes are located in different genomic regions. Mutations in this gene have been associated with paragangliomas. Alternatively spliced transcript variants encoding different isoforms have been described.
Mouse Succinate dehydrogenase cytochrome b560 subunit, mitochondrial (SDHC) ELISA Kit employs a two-site sandwich ELISA to quantitate SDHC in samples. An antibody specific for SDHC has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySDHC present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SDHC is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SDHC bound in the initial step. The color development is stopped and the intensity of the color is measured.
