The epithelial cell adhesion molecule (EpCAM, CD326) is a glycoprotein of ~40 kd that was originally identified as a marker for carcinoma, attributable to its high expression on rapidly proliferating tumors of epithelial origin. Normal epithelia express EpCAM at a variable but generally lower level than carcinoma cells. In early studies, EpCAM was proposed to be a cell-cell adhesion molecule.However, recent insights revealed a more versatile role for EpCAM that is not limited only to cell adhesion but includes diverse processes such as signaling, cell migration, proliferation, and differentiation. Cell surface expression of EpCAM may actually prevent cell-cell adhesion.
Human Epithelial cell adhesion molecule (EPCAM) ELISA Kit employs a two-site sandwich ELISA to quantitate EPCAM in samples. An antibody specific for EPCAM has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyEPCAM present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for EPCAM is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of EPCAM bound in the initial step. The color development is stopped and the intensity of the color is measured.
