Dentin sialophosphoprotein, is the only protein produced uniquely by odontoblasts, the cells that produce tooth dentin. It is a non-collagenous SIBLING protein that is later cleaved into three functional proteins: dentin phosphoprotein (also known as phosphophoryn), taken from the C-terminal end, dentin sialoprotein from the N-terminal end, and dentin glycoprotein from the middle of the molecule. Their distribution in the collagen matrix of the forming dentin suggests these proteins play an important role in the regulation of mineral deposition. Additional evidence for this correlation is phenotypically manifested in patients with mutant forms of dentin sialophosphoprotein. Such patients suffer dental anomalies including type III dentinogenesis imperfecta.
Human Dentin sialophosphoprotein (DSPP) ELISA Kit employs a two-site sandwich ELISA to quantitate DSPP in samples. An antibody specific for DSPP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDSPP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DSPP is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DSPP bound in the initial step. The color development is stopped and the intensity of the color is measured.
