Endothelin converting enzyme 1, also known as ECE1, is a enzyme which in humans is encoded by the ECE1 gene.Endothelin-converting enzyme-1 is involved in the proteolytic processing of endothelin-1 (EDN1), -2 (EDN2), and -3 (EDN3) to biologically active peptides.Schmidt et al. (1994) purified a membrane-bound protease activity from bovine endothelial cells that specifically converts the inactive form to EDN1. The enzyme was cleaved with trypsin and peptide sequencing analysis confirmed it to be a zinc-chelating metalloprotease containing the typical HEXXH (HELTH) motif. RT-PCR and cDNA screens were used to isolate the complete cDNAs of the bovine and human enzymes.
Human Endothelin-converting enzyme 1 (ECE1) ELISA Kit employs a two-site sandwich ELISA to quantitate ECE1 in samples. An antibody specific for ECE1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyECE1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ECE1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ECE1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
