Ephrin-A5 prevents axon bundling in cocultures of cortical neurons with astrocytes, a model of late stage nervous system development and differentiation. The EPH and EPH-related receptors comprise the largest subfamily of receptor protein-tyrosine kinases and have been implicated in mediating developmental events, particularly in the nervous system. EPH receptors typically have a single kinase domain and an extracellular region containing a Cys-rich domain and 2 fibronectin type III repeats. The ephrin ligands and receptors have been named by the Eph Nomenclature Committee. Based on their structures and sequence relationships, ephrins are divided into the ephrin-A class, which are anchored to the membrane by a glycosylphosphatidylinositol linkage, and the ephrin-B class, which are transmembrane proteins.
Human Ephrin-A5 (EFNA5) ELISA Kit employs a two-site sandwich ELISA to quantitate EFNA5 in samples. An antibody specific for EFNA5 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyEFNA5 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for EFNA5 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of EFNA5 bound in the initial step. The color development is stopped and the intensity of the color is measured.
