Interleukin 17A (IL17A) is a proinflammatory cytokine secreted by activated T-lymphocytes. It is a potent inducer of the maturation of CD34-positive hematopoietic precursors into neutrophils. The transmembrane protein encoded by IL17RA is a ubiquitous type I membrane glycoprotein that binds with low affinity to interleukin 17A. Interleukin 17A and its receptor play a pathogenic role in many inflammatory and autoimmune diseases such as rheumatoid arthritis. Like other cytokine receptors, this receptor likely has a multimeric structure. Alternative splicing results in multiple transcript variants encoding different isoforms.
Human Interleukin-17 receptor A (IL17RA) ELISA Kit employs a two-site sandwich ELISA to quantitate IL17RA in samples. An antibody specific for IL17RA has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyIL17RA present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for IL17RA is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of IL17RA bound in the initial step. The color development is stopped and the intensity of the color is measured.
