The mucin genes encode epithelial glycoproteins, some of which are secreted and some membrane bound. Each of the genes contains at least one large domain of tandemly repeated sequence that encodes the peptide sequence rich in serine and/or threonine residues, which carries most of the O-linked glycosylation.
Human Mucin-3A (MUC3A) ELISA Kit employs a two-site sandwich ELISA to quantitate MUC3A in samples. An antibody specific for MUC3A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMUC3A present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MUC3A is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MUC3A bound in the initial step. The color development is stopped and the intensity of the color is measured.
