AMSBIO: 3D Culture Cell Harvesting Kit

The AMSBIO Cultrex® 3D Culture Cell Harvesting Kit is based on a non-enzymatic approach that prevents biochemical degradation during processing and overcomes the problems associated with extracting cells from 3D culture in BME or Laminin I. The quality of the harvested cells is ideal for Western blotting and RNA and DNA analysis. In 3D culture, cells are grown in a microenvironment of specialized proteins that mimics in vivo conditions and allows the cells to organise into complex structures rather than uniform monolayers.

This new kit provides an optimised and standardised solution for the isolation and normalization of cell lysates from 3D Culture Matrix, BME or Laminin I for subsequent biochemical analysis.

3D Cultures exhibit cellular behaviours and morphologies similar to those seen in vivo; however, the adaptation of these models for studying biochemical processes has been impeded by the challenge of separating intact or 3D grown cells from extracellular proteins comprising the 3D matrix. Commonly, proteases are employed to degrade these extracellular proteins; however, proteases also degrade proteins on the cell surface, and protease activity may carry over into lysate preparations. Non-enzymatic methodologies have also been described for depolymerising extracellular matrix proteins, although the implementation of these protocols remains problematic for some researchers.
