Easy, fast and reliable PCR detection of Mycoplasma from cell culture

Biotest’s MMB Mycoplasma PCR Kit offers robust detection of Mycoplasma species from cell culture, including the 9 listed in the European Pharmacopeia.

The MMB Mycoplasma PCR kit consists of a primer mixture that guarantees detection of more than 70 Mycoplasma species. All nine Mycoplasma species listed in the European Pharmacopeia are detected with a sensitivity of 50 colony forming units (cfu) or less per ml cell culture supernatant.

Robustness – Broad detection of Mycoplasma

The MMB Mycoplasma PCR Kit consists of a PCR primer mixture which guarantees the detection of all 9 Mycoplasma species listed in the European Pharmacopeia with a sensitivity of at least 50 colony forming units (cfu)/ml cell culture supernatant: Acholeplasma laidlawii, Mycoplasma orale, Mycoplasma arginini, Mycoplasma pneumoniae, Mycoplasma fermentans, Mycoplasma synoviae, Mycoplasma gallisepticum, Spiroplasma citri and Mycoplasma hyorhinis. Phylogenetically closely related microorganisms, e.g. Streptococcus sp., Clostridium sp. and Lactobacillus sp. are not detected.

Extreme sensitivity – Fewer than 50 cfu/ml

The MMB Mycoplasma PCR Kit is specially optimized for highly sensitive and specific detection of Mycoplasma 16S rDNA. Beside the amplification of a specific 260 bp Mycoplasma fragment, an

internal inhibition control with a size of 160 bp is included. Extreme sensitivity below 50 cfu/ml cell culture is obtained with the ready-to-use PCR Master Mix using a Hot Start Taq DNA Polymerase. PCR detection can be performed on any thermal cycler.

Speed – Detection in less than 3.5 hours

Fast sample preparation in under 10 min: The sample preparation consists of three simple steps including centrifugation of the cell culture at maximum speed for 5 minutes followed by resuspension of the Mycoplasma pellet in water. After heating the sample for 3 minutes at 95°C it can immediately be used for PCR.

PCR detection with ready-to-use PCR master mix: The ready-to-use PCR master mix contains all necessary ingredients for the PCR: primers, buffers, nucleotides and internal control reagents. By adding Hot Start Taq DNA Polymerase and the sample DNA or provided Positive Control the PCR reaction can be started. Depending on the thermal cycler the PCR reaction takes up to 2 hours.

Ready-to-load PCR buffer for immediate gel loading: The provided PCR master mix contains a ready-to-load PCR buffer. The addition of a special loading buffer is not required. For the analysis, a direct transfer of 10 μl PCR reaction on an agarose gel, followed by gel electrophoresis allows the visual inspection for the absence or presence of the desired fragments.
