EED-226 is a novel allosteric Polycomb repressive complex 2 (PRC2) inhibitor (IC50 = 23.4 nM when the substrate is H3K27me0 peptide and IC50 = 53.5 nM when the substrate is mononucleosom). EED-226 induces a conformational change via binding EED which invalidates PRC2 activity. PRC2 plays pivotal roles in transcriptional regulation.
