Microtubule-associated protein 4is a major non-neuronal microtubule-associated protein. This protein contains a domain similar to the microtubule-binding domains of neuronal microtubule-associated protein (MAP2) and microtubule-associated protein tau (MAPT/TAU). This protein promotes microtubule assembly, and has been shown to counteract destabilization of interphase microtubule catastrophe promotion. Cyclin B was found to interact with this protein, which targets cell division cycle 2 (CDC2) kinase to microtubules.The phosphorylation of this protein affects microtubule properties and cell cycle progression. Multiple alternatively spliced transcript variants encoding distinct isoforms have been observed, the full-length nature of three of which are supported.
Rat Microtubule-associated protein 4 (MAP4) ELISA Kit employs a two-site sandwich ELISA to quantitate MAP4 in samples. An antibody specific for MAP4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMAP4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MAP4 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MAP4 bound in the initial step. The color development is stopped and the intensity of the color is measured.
