Presenilin-2 is a protein that is encoded by the PSEN2 gene. Alzheimer's disease (AD) patients with an inherited form of the disease carry mutations in the presenilin proteins (PSEN1; PSEN2) or the amyloid precursor protein (APP). These disease-linked mutations result in increased production of the longer form of amyloid-beta (main component of amyloid deposits found in AD brains). Presenilins are postulated to regulate APP processing through their effects on gamma-secretase, an enzyme that cleaves APP. Also, it is thought that the presenilins are involved in the cleavage of the Notch receptor, such that that they either directly regulate gamma-secretase activity or themselves are protease enzymes. Two alternative transcripts of PSEN2 have been identified. In melanocytic cells PSEN2 gene expression may be regulated by MITF.

Pig Presenilin 2 (PSEN2) ELISA Kit employs a two-site sandwich ELISA to quantitate PSEN2 in samples. An antibody specific for PSEN2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPSEN2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PSEN2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PSEN2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
