Mmp12 encodes a member of the matrix metalloproteinase family of extracellular matrix-degrading enzymes that are involved in tissue remodeling, wound repair, progression of atherosclerosis and tumor invasion. The encoded preproprotein undergoes proteolytic processing to generate a mature, zinc-dependent endopeptidase enzyme. Mice lacking the encoded protein have a diminished capacity to degrade extracellular matrix components, do not develop emphysema in response to long-term exposure to cigarette smoke, and exhibit impaired clearance and increased mortality upon bacterial infection. Mmp12 is located in a cluster of other matrix metalloproteinase genes on chromosome 9. Alternate splicing generates multiple transcript variants encoding distinct isoforms.
Mouse Macrophage metalloelastase (MMP12) ELISA Kit employs a two-site sandwich ELISA to quantitate MMP12 in samples. An antibody specific for MMP12 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMMP12 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MMP12 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MMP12 bound in the initial step. The color development is stopped and the intensity of the color is measured.
