Mmp13 encodes a member of the matrix metalloproteinase family that plays a role in wound healing, skeletal development and bone remodeling. The encoded protein is activated by the removal of an N-terminal activation peptide to generate a zinc-dependent endopeptidase enzyme that can cleave various native collagens, including types I - IV, X and XIV. Mice lacking the encoded protein display profound defects in growth plate cartilage as well as a delay in the endochondral bone development. Lack of the encoded protein also impairs the wound healing process due to reduced keratinocyte migration and vascular density at the wound site. Mmp13 is located in a cluster of other matrix metalloproteinase genes on chromosome 9.
Mouse Matrix metalloproteinase 13 (MMP-13) ELISA Kit employs a two-site sandwich ELISA to quantitate MMP13 in samples. An antibody specific for MMP13 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMMP13 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MMP13 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MMP13 bound in the initial step. The color development is stopped and the intensity of the color is measured.
