Keratin 4 isa member of the keratin gene family. The type II cytokeratins consist of basic or neutral proteins which are arranged in pairs of heterotypic keratin chains coexpressed during differentiation of simple and stratified epithelial tissues. This type II cytokeratin is specifically expressed in differentiated layers of the mucosal and esophageal epithelia with family member KRT13.Mutations in these genes have been associated with White Sponge Nevus, characterized by oral, esophageal, and anal leukoplakia. Human Keratin, type II cytoskeletal 4 (KRT4) ELISA Kit employs a two-site sandwich ELISA to quantitate KRT4 in samples. An antibody specific for KRT4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyKRT4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for KRT4 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of KRT4 bound in the initial step. The color development is stopped and the intensity of the color is measured.
