CDK-activating kinase assembly factor MAT1 encoded by MNAT1, along with cyclin H and CDK7, forms the CDK-activating kinase (CAK) enzymatic complex. This complex activates several cyclin-associated kinases and can also associate with TFIIH to activate transcription by RNA polymerase II. Two transcript variants encoding different isoforms have been found for MNAT1.
Human CDK-activating kinase assembly factor MAT1 (MNAT1) ELISA Kit employs a two-site sandwich ELISA to quantitate MNAT1 in samples. An antibody specific for MNAT1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMNAT1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MNAT1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MNAT1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
