MUC7 encodes a small salivary mucin, which is thought to play a role in facilitating the clearance of bacteria in the oral cavity and to aid in mastication, speech, and swallowing. The central domain of this glycoprotein contains tandem repeats, each composed of 23 amino acids. This antimicrobial protein has antibacterial and antifungal activity. The most common allele contains 6 repeats, and some alleles may be associated with susceptibility to asthma. Alternatively spliced transcript variants with different 5' UTR, but encoding the same protein, have been found for MUC7.
Human Mucin-7 (MUC7) ELISA Kit employs a two-site sandwich ELISA to quantitate MUC7 in samples. An antibody specific for MUC7 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMUC7 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MUC7 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MUC7 bound in the initial step. The color development is stopped and the intensity of the color is measured.
