Agenda
•13:30-13:40 Introduction
•13:40-14:30 A De Novo Antibody Fab Library Using Phage pIX Display
Dr. Lei Shi, Senior Director, Antibody Discovery, RuiYi
Conventional phagemid display uses pIII protein as fusion partner and has been proven to be a very useful tool for antibody discovery. The presentation will discuss the design, construction, and validation of a synthetic antibody Fab library based on phage pIX display.
•14:30-15:20 HTS for Early Stage Antibody Discovery (TBC)
Daniel Wu, Investigator, Institute for Biomedical Research, Novartis (TBC)
Therapeutic antibody generation is a completely different area comparing with reagent antibody. Antibody pool for therapeutic purpose requires more diversified CDR sequences, different epitopes, higher affinities, cross-species activities and even developability potential. Traditional hybridoma technology can hardly meet these criteria. Several key technologies were established to achieve the goal with very limited manpower. More clones can be generated by a modified electrofusion technology. More plates can be handled by a highly automatic hybridoma culture system. And higher screening throughput can be achieved by a homogeneous screening instrument. The instructor will introduce a hybridoma platform and mainly focus on how to setup homogeneous assay for high throughput primary screening.
•15:20-15:40 Coffee Break
•15:40-16:30 Bioanalytical Concerns of Antibody Therapeutics in Preclinical and Clinical Development
Dr. Yongmin Yang, Senior Director, Immunochemistry Bioanalytical Services, WuXi AppTec
Pharmacokinetics (PK), Pharmacodynamics (PD), and immunogenicity are essential elements in the development of antibody therapeutics. This presentation is intended to provide an introduction on bioanalytical aspects of biologics and therapeutic antibodies in particular to scientists and managements working in antibody therapeutics as well as biopharmaceutical sciences in general. The challenges involving in the bioanalytical method development and method validation will be discussed. The presentation will also introduce recent development in bioanalytical guidances related to biologics as well as biosimilars both in China and major parts of the world.
