The 3rd Advanced Training Workshop in the Technology and Applications of Proteomics
 
Proteomics is revealing the secret of life and has found wide applications in all disciplines of life science. It is becoming the frontier and breakthrough point in life science and biotechnology in the 21st century. To further facilitate the development of proteomics in China, Beijing Proteome Research Center (BPRC), State key Laboratory of Proteomics (SKLP) and China Human Proteome Organization (CNHUPO) will hold jointly the 3rd Advanced Training Workshop in the Technology and Applications of Proteomics from July 19 to 22, 2011 in Zhongguancun Life Science Park.
 
BPRC was founded in 2004. It hosts the main body of the SKLP and is the executive headquarter of the International Human Liver Proteome project (HLPP). BPRC is the leader of Chinese proteomics research and has established an internationally renowned research platform for modern proteomics. Many national projects of the 973 and 863 Plans, the Chinese human liver proteome project, and projects funded by the National Natural Science Foundation (in total of more than 100 projects) have been and are being carried out in BPRC.
 
This training workshop will be taught by experts from BPRC who have been working on the forefront of proteomics research. Lectures on “Technology and Methodology for Proteomics, Technology know-how and Experimental Design, Frontiers and Hot Topics in Proteomics Research” will be given on July 19 and 20. On July 21 and 22, trainees will watch live demonstrations of key proteomics techniques. A certificate of completion of the training in proteomics will be issued for all participants.

Experimental Demonstration (half a day per item)
 
1) 2DE-DIGE
Different protein samples are labeled with fluorescent dyes of two different colors so that they can run together in gel electrophoresis. This reduces the experimental error, increases sensitivity and the dynamic range of the detection. The software package can automatically detect proteins of differential expression. This method is suited for differential proteomics of multiple samples with large differences in protein expression among individual samples.
 
2) Techniques in Chromatography
Analytical high efficiency liquid chromatography, capillary high efficiency liquid chromatography, and their in-line use with mass spectrometry will be discussed. Emphases will be given on the components of liquid chromatography, the basic functions of the various components, basic operation and handling of the liquid chromatography, potential problems and trouble shooting in the operation of liquid chromatography. After the lecture, you should have a basic understanding of liquid chromatography and its use in separation of peptides and proteins.
 
3) Operation of a MALDI TOF/TOF Mass Spectromter
We will teach you how to: desalt the sample, prepare the matrix solution, apply samples, set parameter and check hardware, generate and edit the sample table, control the laser, manually acquire data, automatically acquire data in a batch mode, calibrate the instrument, smoothen and correct background, label peaks, use the DataExplore software, operate the mass spectrometer, export data, maintain database, and perform routine instrument maintenance.
 
After the session, you should have a basic idea of how to use MALDI-TOF/TOF mass spectrometry.
 
4) Protein identification with an in-line liquid chromatography electro-spray mass spectrometry (LC-ESI-MS)
 
The technique of LC-ESI-MS/MS is the method of choice for the identification of proteins. It is well suited for complex mixture of peptides. The working horse in the field is capillary reverse phase liquid chromatography, coupled with a Nano-spray source, in-line with a liner Ion Trap mass spectrometer (LTQ). Such combination has proven to be efficient and accurate, and it can be used to identify proteins continuously with high sensitivity. We will cover the keys to optimized protein identification techniques in this session: the condition for best HPLC separation and parameter settings for mass spectrometry, as well as parameter settings for data base search.
