| Cell Line Description |
Saos-2 (osteosarcoma) is a cell line that was isolated by Fogh et al. in 1973 from a primary osteosarcoma in an 11-year-old Caucasian girl. Saos-2 cells express the 1,25-dihydroxyvitamin D3 receptor and have high basal alkaline phosphatase activity. They express the parathyroid hormone (PTH) receptor and produce cyclic adenosine monophosphate after PTH treatment. These cells do not form tumors when injected subcutaneously into immunocompromised mice. However, when injected into diffusion chambers implanted intraperitoneally in immunocompromised mice, Saos-2 cells produce mineralized matrix, a defining characteristic of osteoblasts. These cells have become a widely accepted model for studying osteosarcoma and bone biology due to their osteogenic properties and ability to produce a bone-like extracellular matrix. Saos-2 cells also exhibit an aneuploid karyotype, lacking several chromosomes but having extra copies of other chromosomes, which is typical of many cancer cell lines. They are negative for mycoplasma and have a robust calcification capacity, making them suitable for use in assays related to mineral deposition. |
