The protein encoded by the DPPIV gene is an antigenic enzyme expressed on the surface of most cell types and is associated with immune regulation, signal transduction and apoptosis. It is an intrinsic membrane glycoprotein and a serine exopeptidase that cleaves X-proline dipeptides from the N-terminus of polypeptides.DPPⅣ plays a major role in glucose metabolism. It is responsible for the degradation of incretins such as GLP-1.Furthermore it appears to work as a suppressor in the development of cancer and tumours.CD26/DPPIV plays an important role in tumor biology, and is useful as a marker for various cancers, with its levels either on the cell surface or in the serum being increased in some neoplasms and decreased in others.
Rat Dipeptidyl peptidase 4 (DPP4) ELISA Kit employs a two-site sandwich ELISA to quantitate DPP4 in samples. An antibody specific for DPP4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDPP4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DPP4 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DPP4 bound in the initial step. The color development is stopped and the intensity of the color is measured.
