MMP7 encodes a member of the peptidase M10 family of matrix metalloproteinases (MMPs). Proteins in this family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. The encoded preproprotein is proteolytically processed to generate the mature protease. This secreted protease breaks down proteoglycans, fibronectin, elastin and casein and differs from most MMP family members in that it lacks a conserved C-terminal hemopexin domain. The enzyme is involved in wound healing, and studies in mice suggest that it regulates the activity of defensins in intestinal mucosa. MMP7 is part of a cluster of MMP genes on chromosome 11. MMP7 exhibits elevated expression levels in multiple human cancers.
Rat Matrix metalloproteinase 7 (MMP7) ELISA Kit employs a two-site sandwich ELISA to quantitate MMP7 in samples. An antibody specific for MMP7 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMMP7 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MMP7 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MMP7 bound in the initial step. The color development is stopped and the intensity of the color is measured.
