At a concentration of 10^-12 M, PAF causes life threatening inflammation of the airways to induce asthma like symptoms.Toxins such as fragments of destroyed bacteria induce the synthesis of PAF, which causes a drop in blood pressure and reduced volume of blood pumped by the heart, which leads to shock and maybe death.
Rat Platelet activating factor (PAF) ELISA Kit employs a two-site sandwich ELISA to quantitate PAF in samples. An antibody specific for PAF has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPAF present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PAF is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PAF bound in the initial step. The color development is stopped and the intensity of the color is measured.
