PER2 is a member of the Period family of genes and is expressed in a circadian pattern in the suprachiasmatic nucleus, the primary circadian pacemaker in the mammalian brain. Genes in this family encode components of the circadian rhythms of locomotor activity, metabolism, and behavior. Circadian expression in the suprachiasmatic nucleus continues in constant darkness, and a shift in the light/dark cycle evokes a proportional shift of gene expression in the suprachiasmatic nucleus. The specific function of this gene is not yet known.A new genetic test from a cheek swab can use Per2 expression levels to tell whether a person is an early morning person or a "night owl". It has also been shown that Per2 and Bmal1 work in opposition to each other.
Rat Period circadian protein homolog 2 (PER2) ELISA Kit employs a two-site sandwich ELISA to quantitate PER2 in samples. An antibody specific for PER2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPER2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PER2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PER2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
