This protein is downregulated in prediabetic and type 2 diabetes mellitus patients. Certain allelic variations in this gene increase the risk of the development of obesity.Ppargc1b encodes a predicted 1,014-amino acid protein, and human and mouse PPARGC1B share 70% amino acid sequence identity. Ppargc1b contains 3 N-terminal LXXLL motifs, 2 glutamic/aspartic acid-rich acidic domains, a binding site for host cell factor (HCF1), and a C-terminal RNA recognition motif.

After removing any unbound substances, a biotin-conjugated antibody specific for PPARGC1B is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PPARGC1B bound in the initial step. The color development is stopped and the intensity of the color is measured.
