The protein encoded by TNFsR-II is a member of the TNF-receptor superfamily. This protein and TNF-receptor 1 form a heterocomplex that mediates the recruitment of two anti-apoptotic proteins, c-IAP1 and c-IAP2, which possess E3 ubiquitin ligase activity. The function of IAPs in TNF-receptor signalling is unknown, however, c-IAP1 is thought to potentiate TNF-induced apoptosis by the ubiquitination and degradation of TNF-receptor-associated factor 2, which mediates anti-apoptotic signals. Knockout studies in mice also suggest a role of this protein in protecting neurons from apoptosis by stimulating antioxidative pathways. [provided by RefSeq, Jul 2008]
Rat Tumor necrosis factor soluble receptor II (TNFsR-II) ELISA Kit employs a two-site sandwich ELISA to quantitate TNFsR-II in samples. An antibody specific for TNFsR-II has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTNFsR-II present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TNFsR-II is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TNFsR-II bound in the initial step. The color development is stopped and the intensity of the color is measured.
