Alkaline phosphatase (ALP, ALKP, ALPase, Alk Phos) (EC 3.1.3.1) or basic phosphatase is a homodimeric protein enzyme of 86 kilodaltons, containing two zinc atoms crucial to its catalytic function per monomer, and is optimally active at alkaline pH environments. As its name indicates, ALP functions best under alkaline pH environments and has the physiological role of dephosphorylating compounds. The enzyme is found across a plethora of organisms, prokaryotes and eukaryotes alike, with the same general function but in different structural forms suitable to the environment they function in.
Rabbit Total alkaline phosphatase (TALP) ELISA Kit employs a two-site sandwich ELISA to quantitate TALP in samples. An antibody specific for TALP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTALP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TALP is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TALP bound in the initial step. The color development is stopped and the intensity of the color is measured.
