Myeloperoxidase (MPO) is a peroxidase enzyme that in humans is encoded by the MPO gene on chromosome 17. MPO is most abundantly expressed in neutrophil granulocytes (a subtype of white blood cells), and produces hypohalous acids to carry out their antimicrobial activity. It is a lysosomal protein stored in azurophilic granules of the neutrophil and released into the extracellular space during degranulation. MPO has a heme pigment, which causes its green color in secretions rich in neutrophils, such as pus and some forms of mucus.

Pig Myeloperoxidase (MPO) ELISA Kit employs a two-site sandwich ELISA to quantitate MPO in samples. An antibody specific for MPO has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMPO present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MPO is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MPO bound in the initial step. The color development is stopped and the intensity of the color is measured.
