DEPDC5 encodes a member of the IML1 family of proteins involved in G-protein signaling pathways. The mechanistic target of rapamycin complex 1 (mTORC1) pathway regulates cell growth by sensing the availability of nutrients. The protein encoded by DEPDC5 is a component of the GATOR1 (GAP activity toward Rags) complex which inhibits the amino acid-sensing branch of the mTORC1 pathway. Mutations in DEPDC5 are associated with autosomal dominant familial focal epilepsy with variable foci. A single nucleotide polymorphism in an intron of DEPDC5 has been associated with an increased risk of hepatocellular carcinoma in individuals with chronic hepatitis C virus infection. Alternative splicing results in multiple transcript variants.
Mouse DEP domain-containing protein 5 (DEPDC5) ELISA Kit employs a two-site sandwich ELISA to quantitate DEPDC5 in samples. An antibody specific for DEPDC5 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDEPDC5 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DEPDC5 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DEPDC5 bound in the initial step. The color development is stopped and the intensity of the color is measured.
