6-Keto Prostaglandin F1α (6-k-PGF1α) is a stable hydrolyzed product of unstable Prostacyclin (PGI2) which is derived from Prostaglandin H2 (PGH2), which in turn is synthesized from the arachidonic pathway. Through the arachidonic pathway, it is the major product produced by the endothelial cells, although it is also synthesized in other cell types such as macrophages. PGI2 inhibits platelet aggregation and induces vasodilation. Quantitation of PGI2 production can be made by determining the level of 6-k-PGF1α.

Standards and samples are pipetted into the wells and any6ketoPGF1a present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for 6ketoPGF1a is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of 6ketoPGF1a bound in the initial step. The color development is stopped and the intensity of the color is measured.
