N-terminal sequencing
Principle:
The N-terminal sequencing is based on Edman degradation. There are three steps in each cycle: Coupling of PITC with the amino-terminal residue, Cleavage of the amino-terminal residue via cyclization in acidic media, and conversion of the ATZ derivative formed to the more stable PTC derivative, which may be identified by chromatography

Equipments: ABI491A protein sequencer
Samples Requirements:
1. Protein samples could be in the forms of dry powder, aqueous or PVDF membrane (if on membrane, protein should be less than 40KDa and stained with Ponceaus)
2. Protein amount and purity: amount>10pmol; purity>90%; concentration>1mg/ml
3. Salt concentration: Volatile salt<20mM, nonvolatile salt<5mM,Tris buffer is refusedused
4. Please inform us of the scope of the Molecular Weight
