BackgroundThe TSLP receptor (TSLPR, also known as CRLM-2) is a heterodimeric cytokine receptor composed of a TSLP-binding subunit (TSLPRα) and the α-subunit of the IL-7 receptor (IL-7Rα). TSLPR mRNA is expressed in various immune cell types, including dendritic cells (DCs), T cells, B cells, mast cells, NKT cells, and monocytes, as well as in tissues such as the heart, skeletal muscle, kidney, and liver. While TSLPR has a low affinity for TSLP, it forms a high-affinity binding site for TSLP when combined with IL-7Rα, thereby initiating signaling. The TSLPR regulates target gene expression through the JAK/STAT signaling pathway, involving signal transducers and activators of transcription STAT3, STAT5, and STAT1. Genetic, experimental, and clinical evidence indicates that the TSLP-TSLPR pathway is implicated in the pathogenesis of allergic diseases, including atopic dermatitis (AD) and asthma. bio-equip.cn
