BackgroundNCAM1 is a neural adhesion protein (NCAM) that belongs to the immunoglobulin superfamily. Human NCAM-1 is synthesized as a 761 aa preproprecursor, containing a signal sequence, a large GPI-linked extracellular domain (ECD), and a short C-terminal prosegment. The mature ECD contains five C-2 type Ig-like domains and two fibronectin type III domains. The mature human ECD shares 93% and 95% aa sequence identity with the mouse and rat NCAM-1 ECD, respectively. The encoded protein is involved in cell-to-cell interactions as well as cell-matrix interactions during development and differentiation. NCAM1 is involved in neuron-neuron adhesion, neurite fasciculation, the outgrowth of neurites, etc. It has also been shown to be involved in the expansion of T cells and dendritic cells which play an important role in immune surveillance.
